A comparative study for ultrastructural localization of intracellular immunoglobulins using peroxidase conjugates.

چکیده
Fixation with 4% formaldehyde/24 hours, 1% formaldehyde and 1% glutaraldehyde/1-1.5 hours and 1-1.5% glutaraldehyde/1-1.5 hours provided satisfactory ultrastructural conservation of the tissues. None of these fixatives appeared to denature IgG globulins, but formation of cross-linkages prevents conjugates from reaching antigenic sites. Weak aldehyde fixation may enhance penetration of conjugates; however, ultrastructural detail is poorly preserved and proteins will leak out from insufficiently fixed tissue, thus increasing the possibility of staining artifacts.

Ultrastructural detection of alpha1-fetoprotein in hepatomas by use of peroxidase-labelled antibodies.
Hepatomas were induced in rats by feeding N-nitrosomorpholine. Nodules from alpha1-fetoprotein (AFP)-producing hepatoma were taken and subcutaneously injected into syngeneic hosts. From the first generation, the hepatoma producing most AFP was selected for further transfers. Four weeks after transplantation, the AFP concentration in the host rats was in the range of 0.8 to 1.5 mg AFP/ml serum. ...
متن کاملThe ultrastructural localization of immunoglobulins in human b cells of immunoproliferative diseases.
The cellular distribution of immunoglobulins in human malignant and normal B cells was investigated by immunoelectron microscopy by direct incubation of fixed cells with electron microscopy by direct incubation of fixed cells with peroxidase-coupled antibody. These conjugates penetrated into the cell, resulting in the simultaneous detection of surface and cytoplasmic immunoglobulins. The latter...
متن کاملCoupled intracellular horseradish peroxidase-monoamine oxidase histochemistry: description of the technique and its application to the study of physiologically identified tuberomammillary neurons.
A technique is described which couples visualization of intracellular horseradish peroxidase (HRP) with histochemical localization of monoamine oxidase (MAO), permitting simultaneous study of histochemistry, morphology and physiology in a single neuron. Using this technique, individual neurons int he tuberomammillary nucleus of the hypothalamus were stained in a Golgi-like fashion, revealing th...
متن کاملPresence of Immunoglobulins on the Surface of Lesion-Derived Amastigotes of Leishmania major As a Tool for Isolation of Amastigote Stage from BALB/c Mice
The onset of infections by Leishmania parasites mainly caused by amastigote growth inside the macrophages. One of the important properties of lesion-derived amastigote is thought to be the attachment of various host proteins including immunoglobulins on the surface of amastigote. In this study, the presence of immunoglobulins on the surface of lesion-derived amastigotes was detected by Western ...
متن کاملPlasma Membrane and Internalized Immunoglobulins of Lymph Node Cells Studied with Conjugates of Antibody or Its Fab Fragments with Horseradish Peroxidase
Normal rat and mouse lymphoid cells were incubated at 0 degrees -4 degrees C for 1 h with purified rabbit or sheep antirat (mouse) immunoglobulin (Ig)-horseradish peroxidase (PO) conjugates or with Fab fragments of antibody coupled with peroxidase. Cells were subsequently washed and incubated in fresh medium, without labeled antibody or Fab fragments for 5-30 min at 20 degrees or 37 degrees C. ...
